Biological Background: PRMT2 Function and Localization
- Protein arginine N-methyltransferase 2 (PRMT2), also known as histone-arginine N-methyltransferase PRMT2, is encoded by the PRMT2 gene (alternative names: HMT1, HRMT1L1). It belongs to the protein arginine methyltransferase family.
- Functions as an arginine methyltransferase that methylates the guanidino nitrogens of arginyl residues in proteins such as STAT3, FBL, and histone H4. Related references: PMID:19405910
- Acts as a transcriptional coactivator by interacting with nuclear receptors: it enhances androgen receptor (AR)-mediated transactivation with NCOA2, and estrogen receptor (ER)-mediated transactivation with estrogen. - For Western blotting, PRMT2 has a predicted molecular weight of approximately 49 kDa. Consider using whole cell lysates from human cell lines such as LNCaP (prostate) or MCF-7 (breast), which express PRMT2, and validate with appropriate controls.
- In ELISA applications, optimize coating conditions and antibody dilutions; cross-reactivity with other PRMT family members has not been tested.
