Biological Background: PRKG2 Function and Localization
- cGMP-dependent protein kinase 2 (cGKII) is a serine/threonine-protein kinase encoded by the PRKG2 gene; alternative splicing generates multiple isoforms.
- Acts as a crucial regulator of intestinal secretion by phosphorylating and activating CFTR on the plasma membrane, mediating cGMP-dependent translocation of CFTR in jejunum. Related references: PMID:33106379
- In neurons, acts downstream of NMDAR to promote plasma membrane accumulation of GRIA1/GLUR1 at synapses, enhancing synaptic plasticity by phosphorylating GRIA1 at Ser-863 (by similarity).
- In mechanically stimulated osteoblasts, activates the MAPK/ERK pathway (MAPK3/ERK1 and MAPK1/ERK2), inducing FOS family transcription factors critical for the anabolic bone response to mechanical stimulation (by similarity).
- Predominantly localized at the apical cell membrane.
- Highly expressed in brain, lung, and intestinal mucosa.
- Contains cGMP-binding and ATP-binding domains; classified as a serine/threonine-protein kinase; associated with disease variants linked to dwarfism.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-160); it may detect full-length PRKG2 or N-terminal-containing isoforms.
- For Western blot (WB), use denaturing conditions; the predicted molecular weight is approximately 87 kDa.
- For immunofluorescence/immunocytochemistry (IF/ICC), the antibody may label apical membranes; validate in intestinal or renal epithelial cell models.
- In immunoprecipitation (IP) and ELISA, include appropriate controls and optimize buffer conditions for your sample type.
