Biological Background: PRKCSH Function and Localization
- PRKCSH encodes glucosidase 2 subunit beta, also known as 80K-H protein, protein kinase C substrate 60.1 kDa protein heavy chain (PKCSH), with aliases GIIB, PCLD, PLD1, G19P1, VASAP-60, and AGE-R2.
- Functions as the regulatory subunit of glucosidase II, which sequentially cleaves alpha-1,3-linked glucose residues from immature glycoproteins in the endoplasmic reticulum, a critical step in N-glycan processing.

PMID:10929008 - Required for proper biogenesis and plasma membrane trafficking of PKD1/polycystin-1 in primary cilia, linking PRKCSH to polycystic liver disease.
- Localized to the endoplasmic reticulum lumen, consistent with its role in glycoprotein quality control and ER-associated degradation.
- Annotated with keywords: calcium-binding, disulfide bond, glycoprotein, phosphoprotein, and alternative splicing, indicating post-translational modifications and functional diversity.
- Belongs to the PRKCSH family; molecular weight of canonical isoform is approximately 59.4 kDa (predicted) and observed at 59 kDa, with potential isoforms from alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal peptide (aa 429-528), so the antibody is expected to recognize full-length PRKCSH (~59kDa on Western blot) and may detect isoforms containing this sequence. Validate with lysates from human cell lines known to express PRKCSH.
- For Western blotting, optimize loading and primary antibody dilution using a positive control such as HEK293T or HeLa cell extracts. Consider using a reducing gel system given the presence of disulfide bonds in PRKCSH.
- In ELISA assays, this antibody can be employed either as a capture or detection reagent; titrate against recombinant PRKCSH protein or peptide to establish dynamic range. Pair with a matched detection antibody if available for sandwich ELISA development.
