Biological Background: PRKCQ Function and Localization
- PRKCQ (nPKC-theta) is a calcium-independent, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine-protein kinase that plays a non-redundant role in T-cell receptor (TCR) signaling, including T-cell activation, proliferation, differentiation, and survival.
- In TCR/CD28-co-stimulated T-cells, PRKCQ is required for the activation of NF-kappa-B and JUN, which drive IL2 production, and it participates in calcium-dependent NFATC1 and NFATC2 transactivation. Related references: PMID:21964608
- It mediates canonical NF-kappa-B pathway activation by directly phosphorylating CARD11, leading to recruitment of the BCL10-MALT1 complex and IKK activation, and indirectly influences the non-canonical NF-kappa-B pathway.
- During T-cell differentiation, PRKCQ is crucial for the development of T-helper 2 (Th2) cells and pro-inflammatory Th17 cells, with a minor role in Th1 responses.
- In platelets, PRKCQ regulates signal transduction downstream of integrin ITGA2B and thrombin receptors, promoting 'outside-in' signaling and granule secretion.
- PRKCQ phosphorylates a variety of substrates: it phosphorylates CBLB to promote its degradation, BAD to mediate pro-survival signals, and IRS1 to modulate insulin signaling, among others.
- It phosphorylates CCDC88A/GIV, inhibiting its guanine nucleotide exchange factor activity, which affects cell migration and signaling. Related references: PMID:23509302
- Phosphorylates and activates LRRK1, which in turn phosphorylates RAB proteins involved in intracellular trafficking. Related references: PMID:36040231
- The protein localizes to the cytoplasm and cell membrane, consistent with its role in signal transduction.
- PRKCQ is expressed in skeletal muscle, T-cells, megakaryoblastic cells, and platelets, and contains characteristic kinase domains, zinc-finger motifs, and regulatory phosphorylation/ubiquitination sites.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering the N-terminal region (amino acids 1-230) of human PRKCQ. This design may detect full-length protein and potential isoforms; validate with appropriate controls in your sample system.
- For Western blot, the predicted molecular weight is 82 kDa. Use appropriate loading controls and optimize antibody dilution for your specific lysates.
- Cross-reactivity with human, mouse, and rat enables multi-species studies. Perform initial titration experiments in each species to determine optimal working concentrations.
- In ELISA applications, direct or indirect detection methods may be used. Ensure antigen coating and antibody pairing are optimized for your assay format and sensitivity requirements.
- Shipment is on ice bags; upon receipt, store as recommended and minimize freeze-thaw cycles to maintain antibody integrity.
