Biological Background: PRKCQ Function and Localization

  • PRKCQ (nPKC-theta) is a calcium-independent, phospholipid- and diacylglycerol (DAG)-dependent serine/threonine-protein kinase that plays a non-redundant role in T-cell receptor (TCR) signaling, including T-cell activation, proliferation, differentiation, and survival.
  • In TCR/CD28-co-stimulated T-cells, PRKCQ is required for the activation of NF-kappa-B and JUN, which drive IL2 production, and it participates in calcium-dependent NFATC1 and NFATC2 transactivation. Related references: PMID:21964608
  • It mediates canonical NF-kappa-B pathway activation by directly phosphorylating CARD11, leading to recruitment of the BCL10-MALT1 complex and IKK activation, and indirectly influences the non-canonical NF-kappa-B pathway.
  • During T-cell differentiation, PRKCQ is crucial for the development of T-helper 2 (Th2) cells and pro-inflammatory Th17 cells, with a minor role in Th1 responses.
