Biological Background: PRKAG1 Function and Localization
- PRKAG1 encodes the 5'-AMP-activated protein kinase subunit gamma-1, the regulatory gamma subunit of the AMPK heterotrimer, which acts as a cellular energy sensor.
- Acts as the AMP/ATP-binding subunit of AMPK, an energy sensor protein kinase that regulates cellular energy metabolism by activating energy-producing pathways and inhibiting energy-consuming processes. Related references: PMID:21680840 PMID:24563466
- AMP binding to PRKAG1 leads to allosteric activation, phosphorylation, and prevention of dephosphorylation of the catalytic alpha subunit (PRKAA1/PRKAA2), while ATP promotes dephosphorylation and inactivation of AMPK. Related references: PMID:21680840 PMID:24563466
- Involved in the inhibition of fatty acid, cholesterol, and lipid biosynthesis under low intracellular ATP conditions, contributing to metabolic regulation. Related references: PMID:21680840 PMID:24563466
- Also regulates cellular polarity by remodeling the actin cytoskeleton, probably through indirect activation of myosin. Related references: PMID:21680840 PMID:24563466
- Contains two cystathionine-beta-synthase (CBS) domains that form the nucleotide-binding pockets for AMP, ADP, and ATP.
- Classified as a glycoprotein and phosphoprotein and reported to undergo alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen spans the full-length human PRKAG1 (aa 1-331), which may detect all known isoforms; verify in the relevant sample system.
- For Western blot, an approximate band at 38 kDa is expected; use appropriate controls and consider validation in target tissues.
- For IHC-P and IF/ICC, optimization of antigen retrieval and blocking conditions may be necessary; include positive and negative controls.
- In ELISA, this antibody can be used for quantitative detection of PRKAG1; confirm linearity and sensitivity in your assay setup.
- Cross-reactivity with mouse and rat orthologs allows translation of findings to common rodent models; validate expression levels in the chosen model.
