Biological Background: PRK2/PKN2 Function and Localization

  • Encodes serine/threonine-protein kinase N2 (PKN2), also known as PKN gamma, protein kinase C-like 2, and protein-kinase C-related kinase 2; a member of the PKC-related kinase family.
  • Acts as a Rho/Rac effector kinase involved in diverse signal transduction pathways, including regulation of cell cycle progression, actin cytoskeleton assembly, cell migration, cell adhesion, and transcriptional activation.
  • Localizes to multiple subcellular compartments: cytoplasm, nucleus, membrane, lamellipodium, cleavage furrow, midbody, and cell junctions, consistent with its roles in cytokinesis and adhesion.
  • Exhibits ubiquitous expression, with notable expression in various tumor cell lines, particularly in bladder tumor cells.
  • Phosphorylates HDAC5, impairing its nuclear import; phosphorylates CTTN, reducing its actin-binding capacity; and stimulates FYN kinase activity during keratinocyte differentiation.
  • Involved in negative regulation of ciliogenesis (PubMed:27104747). Related references: PMID:27104747
  • During microbial infection, phosphorylates HCV NS5B, stimulating HCV RNA replication.
  • Key post‑translational modifications include phosphorylation and acetylation; the protein contains a coiled‑coil domain and belongs to the kinase superfamily.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 600–700 near the C‑terminus; the antibody may recognize full‑length (~112 kDa) and potential cleavage products. Validate in your target sample system.
  • For Western blot, a predominant band around 112 kDa is expected. Optimize antibody dilution (e.g., 1:500–1:2000) and blocking conditions empirically.
  • For ELISA, perform a titration series to determine the optimal capture/detection antibody concentration for your antigen format.
  • As a polyclonal reagent, lot‑to‑lot variation may occur; verify each new lot with appropriate positive and negative controls.
  • Cross‑reactivity with mouse and rat is reported; if using these species, confirm specificity in your experimental context.
