Biological Background: PRIM2 Function and Localization
- PRIM2 (DNA primase large subunit, also known as p58) is the regulatory subunit of the DNA primase complex and an integral component of the DNA polymerase alpha complex, essential for the initiation of DNA synthesis. Related references: PMID:17893144, PMID:25550159, PMID:26975377
- During S phase, the DNA polymerase alpha complex is recruited to replication forks, where it synthesizes short RNA primers on both the leading and lagging strands. Related references: PMID:17893144
- Within the primase complex, both PRIM1 (catalytic) and PRIM2 (regulatory) subunits are required for the initial di-nucleotide formation, but primer extension depends solely on PRIM1. Related references: PMID:17893144, PMID:25550159
- PRIM2 binds RNA:DNA duplexes and coordinates the switch between primase and polymerase activities by interacting with PRIM1 and POLA2.
- The protein contains a 4Fe-4S iron-sulfur cluster and is subject to phosphorylation, suggesting regulatory roles in DNA replication and cell cycle progression.
- Localized primarily in the nucleus, PRIM2 is active at replication foci and is part of the primosome complex.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–170) of human PRIM2. This region may be involved in protein-protein interactions; consider its accessibility in native versus denaturing conditions.
- For Western blot (WB), a band at approximately 59 kDa is expected in human and mouse cell lysates. Optimize antibody dilution (e.g., 1:500 – 1:2000) and validate with appropriate controls.
- ELISA applications may require coating with the recombinant antigen or cell extracts; the polyclonal nature provides broad epitope coverage, but batch-to-batch variability should be assessed.
