- Proteoglycan 4 (PRG4), also known as lubricin, megakaryocyte-stimulating factor (MSF), and superficial zone proteoglycan (SZP), is encoded by the PRG4 gene. It is a secreted glycoprotein that undergoes alternative splicing, giving rise to multiple isoforms with distinct tissue distributions and functions.
- PRG4 plays a critical role in boundary lubrication within articulating joints, preventing protein deposition onto cartilage from synovial fluid and inhibiting the adhesion of synovial cells to the cartilage surface, thus maintaining joint homeostasis.
- Isoform F of PRG4 acts as a growth factor, stimulating the proliferation of primitive hematopoietic and endothelial cell lineages, indicating a role in megakaryocyte and endothelial biology.
- Subcellularly, PRG4 is secreted into the extracellular space, consistent with its functions in synovial fluid and as a matricellular signal.
- Tissue expression is highest in synovial tissue, cartilage, and liver, with lower levels in heart and lung. Isoform B is expressed in kidney, lung, liver, heart, and brain, while isoforms C and D are widely expressed.

Experimental Guidance and Technical Tips
- The immunogen corresponds to a region in the C-terminal part of human PRG4 (amino acids 1264-1363 of isoform NP_001121180.2). This region may be present in multiple isoforms; verify isoform specificity if required.
- For Western blot, the predicted molecular weight is approximately 151 kDa; however, due to extensive glycosylation, the observed band may migrate at a higher molecular weight. Optimize gel percentage and transfer conditions accordingly.
- For ELISA, the antibody may be used as a capture or detection reagent; pairing with another PRG4 antibody is recommended for sandwich assays.
- This antibody shows cross-reactivity with rat; consider validating in rat synovial fluid or cartilage extracts if using rat models.
