Biological Background: PRG2 Function and Localization
- The PRG2 gene encodes bone marrow proteoglycan, also known as proteoglycan 2, eosinophil major basic protein (MBP) homolog, or proMBP. The protein exists as both an inactive proform and a mature, active secreted form generated through proteolytic processing.
- PRG2 functions as a potent cytotoxin and helminthotoxin, playing a critical role in antiparasitic defense mechanisms. It also induces non-cytolytic histamine release from human basophils, contributing to immune hypersensitivity reactions.
- The proform of PRG2 acts as a proteinase inhibitor, specifically reducing the activity of pregnancy-associated plasma protein-A (PAPPA), and is highly abundant in placenta and pregnancy serum.
- PRG2 is a secreted protein that localizes to cytoplasmic vesicles, particularly secretory vesicles, consistent with its release during eosinophil degranulation and immune responses.
- The protein has been detected in plasma, urine, and placenta, with lower expression in tissues such as kidney, myometrium, endometrium, ovaries, breast, prostate, bone marrow, and colon.
- Post-translational modifications include glycosylation, disulfide bond formation, and nitration, and it contains a heparin-binding domain. These features, along with alternative splicing, contribute to its structural and functional diversity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 17–222 of human PRG2 (NP_002719.3), covering the mature protein region. This design may recognize both proform and mature PRG2 in Western blot, potentially yielding bands around 23–25 kDa as observed.
- For WB, consider using reducing conditions and validate the detected band sizes against predicted molecular weights of the proform (~25 kDa) and mature form (~23 kDa) in relevant human samples such as eosinophil lysates or placental tissue extracts.
- For IF-P, fixation and permeabilization conditions should be optimized; PRG2 is expected in cytoplasmic secretory vesicles. Strong signal may be obtained in tissues with high eosinophil infiltration or placenta.
- ELISA applications may be used for detecting PRG2 in serum, plasma, or urine, but assay conditions (capture/detection) should be validated for the proform and mature form individually if differentiation is required.
