Biological Background: PRG2 Function and Localization
- PRG2 (bone marrow proteoglycan, also known as eosinophil major basic protein, BMPG, MBP1) is a cytotoxin and helminthotoxin. It induces non-cytolytic histamine release from human basophils and participates in antiparasitic defense and immune hypersensitivity reactions.
- The proform of PRG2 acts as a proteinase inhibitor, reducing the activity of pregnancy-associated plasma protein A (PAPPA).
- PRG2 is a secreted protein localized to cytoplasmic vesicles and secretory vesicles.
- Tissue expression: PRG2 is detected in plasma and urine (at protein level) (PubMed:25326458, PubMed:36213313, PubMed:37453717). It is also detected in placenta (PubMed:32337544), with high levels of the proform in placenta and pregnancy serum. In placenta, it localizes to X cells of septa and anchoring villi. - PRG2 is a heparin-binding proteoglycan with a lectin domain. It undergoes post-translational modifications including glycosylation, tyrosine nitration, and disulfide bond formation.
- The protein exhibits antimicrobial and antibiotic activity, contributing to innate immunity.
- Alternative splicing produces multiple isoforms of PRG2.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from amino acids 100–200 of human PRG2. For cross-species applications, verify sequence homology in the target species (reactivity is confirmed for human, mouse, and rat).
- For western blot (WB), the anticipated band is approximately 25 kDa. Prepare appropriate positive and negative controls using cell lines or tissues with known PRG2 expression.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval methods and antibody dilution using a known positive tissue (e.g., placenta, bone marrow).
- For ELISA, consider direct or capture assay formats and validate coating conditions with recombinant PRG2 or native protein from biological samples.
- The polyclonal nature may yield higher sensitivity but requires careful titration to minimize background; test multiple dilutions in the relevant sample system.
