Biological Background: PREX1 Function and Localization

• PREX1 (Phosphatidylinositol 3,4,5-trisphosphate-dependent Rac exchanger 1 protein) is a RAC guanine nucleotide exchange factor (GEF) that activates Rac proteins by exchanging bound GDP for free GTP.
• Its activity is synergistically activated by phosphatidylinositol 3,4,5-trisphosphate (PIP3) and the beta-gamma subunits of heterotrimeric G proteins.
• PREX1 may function downstream of heterotrimeric G proteins in neutrophils, regulating processes such as chemotaxis and oxidative burst.
• The protein is primarily localized in the cytoplasm and cytosol, and associates with the cell membrane upon activation.
• PREX1 is mainly expressed in peripheral blood leukocytes and brain, with intermediate levels in spleen and lymph nodes, and lower expression in other tissues.
