Biological Background: PRDM9 Function and Localization

  • PRDM9 (also known as Meisetz, Hst1, or hybrid sterility protein 1) is a histone methyltransferase that sequentially mono-, di-, and tri-methylates both H3K4 and H3K36 to generate H3K4me3 and H3K36me3, playing a pivotal role in meiotic hotspot determination and recombination. Related references: PMID:16292313 PMID:24095733 PMID:24785241
  • In addition to H3, PRDM9 can mono- and di-methylate histone H4 at Lys20 (H4K20me1/me2) and tri-methylate H3K9, with H3K9me2 being the preferred substrate. Related references: PMID:24785241 PMID:27362481
  • During meiotic prophase, PRDM9 binds specific DNA motifs via its C-terminal zinc finger array, directing local enrichment of H3K4me3 and H3K36me3 on adjacent nucleosomes. Related references: PMID:22028627 PMID:27362481 PMID:29478809
  • This histone modification landscape promotes the formation of programmed DNA double-strand breaks (DSBs) at PRDM9-bound hotspots, initiating meiotic recombination. Related references: PMID:16292313 PMID:29478809
  • PRDM9 interacts with multiple protein complexes during meiosis: it associates with CDYL and EHMT2 in early leptonema, and later recruits EWSR1 and CXXC1, which connect hotspots to the chromosome axis via REC8. Related references: PMID:27932493 PMID:25894966
  • Beyond its role in recombination, PRDM9 functions as a transcriptional activator in early meiotic prophase, leveraging H3K4me3 and H3K36me3 as epigenetic marks for gene activation. Related references: PMID:16292313
  • PRDM9 is subject to automethylation, and its methyltransferase activity can be attenuated by acetylation or phosphorylation of histone H3. Related references: PMID:28126738 PMID:27362481
  • PRDM9 is specifically expressed in germ cells entering meiotic prophase in female fetal gonads and postnatal testis, and localizes to the nucleus and chromosomes. Related references: PMID:16292313 PMID:27932493

Experimental Guidance and Technical Tips

  • This antibody was generated against a recombinant fusion protein spanning amino acids 87–370 of mouse PRDM9, a region that includes the PR/SET domain and excludes the highly repetitive zinc finger array, which may enhance target specificity.
  • It has been reported for use in IHC-P, ELISA, and CUT&Tag applications; consider optimizing antibody titration and assay conditions for your specific model system.
  • Given the observed cross-reactivity with human PRDM9, this antibody may be employed in human sample analysis; however, independent validation in the relevant tissue or cell type is recommended.
  • The predicted molecular weight of PRDM9 is approximately 97 kDa; for Western blotting applications not yet listed, test a panel of positive controls such as meiotic germ cell lysates.
  • For IHC-P, antigen retrieval methods and detection systems should be standardized using tissue sections known to express PRDM9, such as testis or fetal ovary.
