Biological Background: PRC1 Function and Localization
- PRC1 (Protein Regulator of Cytokinesis 1, alias ASE1) is a key mitotic regulator with a predicted molecular mass of 71.6 kDa, encoded by the PRC1 gene (GeneID: 9055). Alternative splicing generates multiple isoforms.
- It functions as a microtubule cross-linking protein, bundling antiparallel microtubules at a spacing of ~35 nm, which is essential for central spindle midzone assembly and successful cytokinesis.
- PRC1 recruits PLK1 to the spindle and is required for KIF14 localization to the central spindle and midbody, thereby promoting downstream signaling (e.g., PLK1-mediated RACGAP1 phosphorylation and ECT2 recruitment) critical for furrow ingression.
- Overexpression of PRC1 has been implicated as an oncogenic driver in bladder cancer, enhancing proliferation, suppressing apoptosis, and facilitating carcinogenesis. Related references: PMID:17409436
- Subcellularly, PRC1 dynamically localizes to the nucleus during interphase, then to the mitotic spindle, spindle midzone, and ultimately the midbody during cytokinesis, also associating with chromosomes.
- Post-translationally, PRC1 is a phosphoprotein and contains coiled-coil domains, consistent with its role in cell cycle-dependent microtubule organization.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal domain (aa 351–620) of human PRC1. This design may recognize multiple isoforms and is predicted to detect endogenous PRC1 in relevant sample systems.
- For Western blotting, a band of approximately 72 kDa is expected. Consider using whole-cell lysates from proliferating cells or synchronized mitotic populations to enhance signal.
- This antibody is validated for IHC-P and IF/ICC. Appropriate fixation and antigen retrieval conditions should be optimized, especially for microtubule-associated targets.
- The oncogenic role of PRC1 in bladder cancer makes it a candidate marker for oncology research; consider validating expression in bladder cancer cell lines or tissue arrays.
