- PRAF2 (JM4/Yip6a) is a member of the PRA1 family proteins involved in intracellular trafficking.
- It may play a role in ER-to-Golgi transport and general vesicular traffic.
- PRAF2 exhibits a proapoptotic function in cerulenin-induced neuroblastoma apoptosis.
- The protein localizes to endosome membranes.
- It is strongly expressed in the brain, particularly in Purkinje cells and cerebellar layers, as well as in small intestine, lung, spleen, and pancreas.
- High expression is observed in neuroblastic tumors and other tumor tissues like breast, colon, lung, and ovary, compared to weaker expression in matched normal tissues.
- It is detected in neuronal cells but not in non-neuronal cells of the cerebral cortex, hippocampus, and lateral ventricles.
- Contains transmembrane helices and is involved in protein transport.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminus (aa 1-50), which may allow detection of full-length PRAF2 and N-terminal variants.
- For Western blot, the predicted molecular weight is ~19 kDa; run a low molecular weight range gel and consider using a positive control from brain or tumor cell lines.
- In ELISA applications, peptide coating or recombinant protein may be used; validate specificity using blocking with the immunogen peptide.
- Cross-reactivity with mouse and rat has been indicated; however, sequence alignment of the immunogen region across species is recommended to confirm expected reactivity.
