- PPP4C encodes the catalytic subunit of serine/threonine-protein phosphatase 4 (PP4), also known as protein phosphatase X (PP-X). It is a member of the PPP family of protein phosphatases.
- PP4 is a multifunctional enzyme involved in critical cellular processes including microtubule organization at centrosomes, maturation of spliceosomal snRNPs, apoptosis, DNA repair, and tumor necrosis factor (TNF)-alpha signaling.
- The PP4 holoenzyme complexes, such as PPP4C-PPP4R1 and PPP4C-PPP4R2-PPP4R3A, regulate histone deacetylation by dephosphorylating HDAC3 and participate in DNA damage response by specifically dephosphorylating gamma-H2AX at Ser-140.
- Subcellular localization includes the cytoplasm, nucleus, and centrosomes, consistent with its roles in cell cycle progression, DNA damage checkpoint signaling, and cytoskeletal organization.
- PPP4C dephosphorylates NDEL1 at CDK1 phosphorylation sites, negatively regulating CDK1 activity during interphase, and dephosphorylates RPA2 to facilitate RAD51 recruitment to chromatin for homologous recombination repair.
- The catalytic activity requires manganese ions as cofactors, and the protein is subject to post-translational modifications including acetylation and methylation.
- Additional functions include activation of c-Jun N-terminal kinase MAPK8, regulation of NF-kappa-B activation, and modulation of cell migration, implicating PP4 in immune and metastatic pathways.
