Biological Background: PPP4C Function and Localization
- PPP4C encodes the catalytic subunit of serine/threonine-protein phosphatase 4 (PP4), also known as protein phosphatase X (PP-X).
- PP4 is a ubiquitous phosphatase involved in numerous cellular processes, including microtubule organization at centrosomes, spliceosomal snRNP maturation, apoptosis, DNA repair, TNF-alpha signaling, JNK activation, histone acetylation regulation, DNA damage checkpoint signaling, NF-kappa-B activation, and cell migration.
- PP4 forms distinct complexes with regulatory subunits such as PPP4R1, PPP4R2, and PPP4R3A, which confer substrate specificity: for example, the PPP4C-PPP4R2-PPP4R3A complex dephosphorylates gamma-H2AX at DNA damage sites, while the PPP4C-PPP4R1 complex may regulate HDAC3.
- The protein localizes predominantly to the cytoplasm and nucleus, with enrichment at the centrosome and microtubule organizing center, consistent with its roles in cytoskeletal regulation and mitosis.
- Post-translational modifications include acetylation and methylation, and the enzyme requires manganese as a cofactor for its hydrolase activity.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering amino acids 1-307 of human PPP4C (NP_002711.1), which represents the full-length catalytic subunit; this design is expected to generate polyclonal antibodies recognizing multiple epitopes.
- For Western blot analysis, a major band near 35 kDa is anticipated; however, post-translational modifications or complex formation may result in additional bands—validate in the relevant sample system.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider that PPP4C localizes to both cytoplasm and nucleus, with centrosomal enrichment; appropriate fixation and permeabilization protocols should be tested.
- For immunoprecipitation (IP) and ELISA applications, standard conditions can be used; note that as a polyclonal preparation, cross-linking to beads or using a bridging antibody may improve capture efficiency.
- Cross-reactivity with mouse and rat orthologs is supported, but sequence alignment and preliminary confirmation in the intended species are recommended before high-throughput assays.
