Biological Background: PPP1R3B Function and Localization

  • Protein phosphatase 1 regulatory subunit 3B (PPP1R3B), also known as GL, PTG, or PPP1R4, is a glycogen-targeting subunit for protein phosphatase 1 (PP1).
  • PPP1R3B facilitates the interaction of PP1 with enzymes of glycogen metabolism, suppressing PP1-mediated dephosphorylation (inactivation) of glycogen phosphorylase and enhancing glycogen synthase activation, thereby limiting glycogen breakdown and promoting glycogen synthesis.
  • Overexpression of PPP1R3B in hepatocytes dramatically increases basal and insulin-stimulated glycogen synthesis (By similarity).
  • PPP1R3B is highly expressed in liver and at lower levels in skeletal muscle (vastus lateralis, gastrocnemius, soleus) at the protein level; mRNA is highest in skeletal muscle, moderate in liver and heart, and weak in placenta and lung.
  • It belongs to the protein phosphatase 1 regulatory subunit family and is a phosphoprotein.
  • Annotations include involvement in carbohydrate and glycogen metabolism, and it has a predicted molecular weight of approximately 33 kDa.

Experimental Guidance and Technical Tips

  • The immunogen (aa 1-140) targets the N-terminus of human PPP1R3B; consider that this region may be conserved across species (cross-reactivity with mouse and rat is reported) and may detect potential isoforms.
  • For Western blot, the predicted molecular weight is ~33 kDa; validate band specificity using positive control lysates from liver or skeletal muscle tissues, taking into account species cross-reactivity.
  • In ELISA applications, optimize coating and detection conditions for quantitative analysis; use recombinant protein or peptide controls to establish assay sensitivity.
  • Note that expression levels may be higher in liver and muscle; consider tissue-specific abundance when planning experiments.
