Biological Background: PPM1E Function and Localization
- PPM1E (Protein phosphatase 1E), also known as POPX1, CaMKP-N, CaMKN, or CAMKPN, is a member of the PP2C family of Ser/Thr protein phosphatases.
- Encoded by the PPM1E gene in humans (GeneID: 22843), it functions as a protein phosphatase that inactivates multifunctional CaM kinases such as CAMK4 and CAMK2.
- Additionally, PPM1E dephosphorylates and inactivates PAK, and may play a role in regulating actin fiber stress breakdown and morphological changes driven by TNK2/CDC42 signaling.
- It also dephosphorylates PRKAA2 (AMPK α2), implicating it in energy-sensing pathways.
- PPM1E localizes to both the nucleus and cytoplasm, indicating a dynamic role in multiple cellular compartments.
- The protein harbors metal-binding sites (Mg²+ or Mn²+) essential for its phosphatase activity, and undergoes alternative splicing.
- Post-translational modifications: identified as a phosphoprotein via proteomics.
- Predicted molecular weight: ~84 kDa, matching the calculated mass of 83952 Da.
Experimental Guidance and Technical Tips
- The immunogen was a synthetic peptide corresponding to amino acids 500–600 of human PPM1E. For Western blot, a band around 84 kDa is expected in human cell lysates. Consider using appropriate positive and negative controls.
- For ELISA applications, optimize coating conditions and antibody concentrations. Validate specificity with recombinant PPM1E or peptide competition assays.
- Note that the antibody is reactive with human samples; cross-reactivity with other species has not been tested. Confirm suitability if working with non-human samples.
- Because PPM1E is a phosphatase that regulates CAMK and PAK pathways, consider stimulating cells to modulate its activity or expression for functional studies.
