Biological Background: PPM1A Function and Localization
- PPM1A (Protein phosphatase 1A), also known as PP2Cα or PP2CA, is a serine/threonine phosphatase encoded by the PPM1A gene in humans. It belongs to the PP2C family of metal-dependent protein phosphatases.
- It functions as a broad-specificity enzyme that negatively regulates TGF-beta signaling by dephosphorylating SMAD2 and SMAD3, leading to their dissociation from SMAD4 and nuclear export, thereby terminating the signal.
- PPM1A plays a critical role in dampening TNF-mediated NF-kappa-B activation by dephosphorylating and inactivating IKBKB/IKKβ kinase.
- It also dephosphorylates AMPK subunits PRKAA1 and PRKAA2, implicating it in energy homeostasis pathways.
- Subcellularly, PPM1A is found in the nucleus, cytoplasm, cytosol, and membrane-associated compartments, consistent with its diverse signaling roles.
- The protein undergoes alternative splicing, is myristoylated, and is itself phosphorylated. It requires magnesium or manganese ions as cofactors for catalytic activity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 161-370) of human PPM1A, which may confer specificity for the canonical PP2Cα isoform; verify performance in the relevant sample system.
- For Western blot (WB), the predicted molecular weight is approximately 42 kDa; consider loading appropriate positive controls (e.g., HeLa or HEK293 lysates) and optimizing blocking conditions.
- For immunofluorescence/ICC, permeabilization and fixation methods may influence detection of nuclear and cytoplasmic pools; validate co-localization markers.
- When used in ELISA, the antibody may be suitable as both capture and detection reagent; perform cross-titration to determine optimal working concentrations.
- As cross-reactivity is confirmed for human, mouse, and rat, species-specific signal should be validated with endogenous tissue extracts or cell lines.
