Biological Background: PPHLN1 Function and Localization
- Periphilin-1 (PPHLN1, also known as CDC7 expression repressor CR, HSPC206, HSPC232, or gastric cancer antigen Ga50) is an RNA-binding protein and core component of the human silencing hub (HUSH) complex.
- PPHLN1 functions within the HUSH complex to epigenetically repress mobile genetic elements, including retroviruses and transposable elements, thereby safeguarding genome integrity. Related references: PMID:15474462, PMID:17963697, PMID:26022416
- The HUSH complex predominantly targets LINE-1 (L1) retrotransposons, where PPHLN1 binds nascent transcripts to initiate silencing and maintain HUSH at chromatin. Related references: PMID:32976585, PMID:39658355
- Through recruitment of the histone methyltransferase SETDB1 and chromatin remodeler MORC2, the HUSH complex promotes H3K9me3 deposition and chromatin compaction at target loci. Related references: PMID:26022416
- PPHLN1 also contributes to HUSH2 complex-mediated repression of interferon-stimulated genes and silencing of unintegrated retroviral DNA. Related references: PMID:30487602, PMID:33144593, PMID:39013473
- Although ubiquitously expressed, PPHLN1 has been implicated in epithelial differentiation and barrier formation. Related references: PMID:12853457
- Subcellularly, PPHLN1 localizes to the nucleus, cytoplasm, and chromosome, consistent with its transcriptional regulatory roles. Post-translational modifications include acetylation and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 195–255 of human PPHLN1; consider sequence alignment with mouse and rat orthologs to assess cross-reactivity specificity.
- For western blot, a band at approximately 53 kDa is expected; validate detection in human, mouse, and rat lysates under reducing conditions.
- This polyclonal antibody is suitable for ELISA; titrate the antibody and include appropriate negative controls to ensure signal specificity.
- When probing HUSH complex components, consider co-immunoprecipitation or chromatin immunoprecipitation workflows to complement functional studies.
