Biological Background: PPARA Function and Localization

  • PPARA is a ligand-activated transcription factor belonging to the nuclear receptor subfamily 1 group C member 1, also known as PPARalpha, NR1C1, or hPPAR.
  • It is a key regulator of lipid metabolism, activated by endogenous ligands such as 1-palmitoyl-2-oleoyl-sn-glycerol-3-phosphocholine (16:0/18:1-GPC) and oleoylethanolamide, a lipid that regulates satiety.
  • Functions as a receptor for peroxisome proliferators, including hypolipidemic drugs and fatty acids, and regulates the peroxisomal beta-oxidation pathway of fatty acids.
  • Transactivation activity requires heterodimerization with the retinoid X receptor alpha (RXRA) and is antagonized by NR2C2.
  • PPARA acts as a transcription activator for genes such as ACOX1 and P450, and may help propagate circadian clock information to metabolic pathways via PER2.
  • Predominantly localized in the nucleus, with tissue-specific expression in skeletal muscle, liver, heart, kidney, and monocytes.
  • Contains conserved zinc-finger DNA-binding domains, metal-binding sites, and is subject to alternative splicing and ubiquitin-like conjugation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a conserved region in human PPARA; cross-reactivity with mouse is reported and should be validated in your experimental system.
  • For Western blot, consider enriching nuclear fractions to enhance detection and include appropriate controls to account for polyclonal antibody variability.
  • The predicted protein weight is 52 kDa; however, post-translational modifications or alternative splicing may result in additional bands.
