Biological Background: PPAP2B Function and Localization
-
PPAP2B, also known as phospholipid phosphatase 3 (PLPP3) or PAP2-beta, is a magnesium-independent lipid phosphate phosphohydrolase encoded by the PLPP3 gene in humans.
-
It catalyzes the dephosphorylation of diverse bioactive lipid phosphate esters including phosphatidate (PA), lysophosphatidate (LPA), diacylglycerol pyrophosphate (DGPP), sphingosine 1-phosphate (S1P), and ceramide 1-phosphate (C1P), as well as N-oleoyl ethanolamine phosphate. Related references: PMID:27694435, PMID:9607309, PMID:9705349
-
The enzyme exhibits both extracellular and intracellular phosphatase activities, enabling hydrolysis and cellular uptake of lipid mediators from the extracellular milieu, thereby regulating signal transduction. Related references: PMID:23591818, PMID:27694435, PMID:9607309
-
Through dephosphorylation of extracellular S1P, PPAP2B plays a critical role in vascular homeostasis, modulating endothelial cell migration, adhesion, survival, proliferation, and pro-inflammatory cytokine production. Related references: PMID:27694435
-
Independently of its phosphatase activity, PPAP2B participates in the Wnt signaling pathway and stabilization of beta-catenin, influencing cell proliferation, migration, and differentiation in angiogenesis and tumor growth, and also plays a role in integrin-mediated cell-cell adhesion. Related references: PMID:20123964, PMID:21569306, PMID:12660161, PMID:16099422
-
Subcellularly, PPAP2B localizes to the plasma membrane, basolateral cell membrane, endoplasmic reticulum, Golgi apparatus, and membrane rafts, reflecting its dynamic trafficking and function in lipid signaling.
-
PPAP2B is ubiquitously expressed, with highest levels in heart and placenta. Related references: PMID:12660161, PMID:9305923
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to the N-terminal region (amino acids 1-100) of human PPAP2B. Consider that the antibody may recognize endogenous PPAP2B across human, mouse, and rat due to high sequence homology in this region.
- In Western blot (WB), PPAP2B is expected to migrate at a molecular weight of approximately 35 kDa. Validate the antibody using positive control lysates from tissues with high expression such as heart or placenta.
- For ELISA, the antibody can be used for detection; optimal working dilution and coating conditions should be determined empirically.
- As a polyclonal antibody, lot-to-lot consistency should be monitored; consider running a standard positive control sample in each experiment.
