Biological Background: PPA1 Function and Localization
- Inorganic pyrophosphatase (PPA1), also known as PPase, PP, PP1, IOPPP, HEL-S-66p, or SID6-8061, is a ubiquitously expressed enzyme that catalyzes the hydrolysis of inorganic diphosphate (PPi) into two orthophosphate molecules, driving biosynthetic reactions and energy metabolism. - Exhibits MAP kinase serine/threonine phosphatase activity toward c-Jun N-terminal kinase (JNK), and in neuronal cells, negatively regulates neurite growth via JNK dephosphorylation.
- Predominantly localized to the cytoplasm, consistent with its role in cellular phosphate homeostasis.
- Member of the inorganic pyrophosphatase family, conserved across eukaryotes and required for multiple biosynthetic pathways.
- Contains acetylation and phosphorylation sites, indicating potential regulation by post-translational modifications.
- Requires magnesium ions as a cofactor for catalytic activity.
Experimental Guidance and Technical Tips
- For Western blot, use whole-cell lysates from human, mouse, or rat tissues/cell lines. The expected band size is approximately 33 kDa.
- In ELISA applications, the immunogen is a full-length recombinant protein; consider optimizing coating conditions and antibody titration for your specific assay format.
- Since PPA1 is widely expressed, validate specificity by including appropriate negative controls (e.g., knockdown lysates or blocking peptide) if off-target binding is a concern.
