Biological Background: Serine/threonine-protein phosphatase PP1-beta catalytic subunit Function and Localization
- Also known as PP1beta, PP-1B, PPP1CD, and PPP1beta, this enzyme is encoded by the PPP1CB gene and belongs to the PP1 family of serine/threonine phosphatases.
- Functions as the catalytic subunit of protein phosphatase 1 (PP1) holoenzymes, which regulate diverse processes including cell division, glycogen metabolism, muscle contractility, and protein synthesis.
- Component of the PTW/PP1 phosphatase complex, which plays a critical role in the control of chromatin structure and the transition from mitosis to interphase.
- In concert with casein kinase I delta and epsilon (CSNK1D and CSNK1E), regulates circadian period length by modulating the phosphorylation dynamics of PER1 and PER2 proteins.
- Dephosphorylates FOXP3 at Ser-418 in regulatory T-cells from rheumatoid arthritis patients, leading to FOXP3 inactivation and Treg dysfunction. Related references: PMID:23396208
- Core component of the SHOC2-MRAS-PP1c (SMP) holophosphatase complex, which specifically dephosphorylates inhibitory sites on RAF1, BRAF, and ARAF kinases to activate the MAPK signaling pathway. Related references: PMID:35768504 PMID:35831509 PMID:36175670
- Localizes to the cytoplasm, nucleus, nucleoplasm, and nucleolus, reflecting its involvement in both cytoplasmic and nuclear signaling events.
- Post-translationally modified by acetylation and phosphorylation, and requires manganese as a cofactor for catalytic activity.
Experimental Guidance and Technical Tips
- The immunogen was a synthetic peptide from amino acids 227-327 of human PP1 beta. For Western blot (WB), expect a band at approximately 37 kDa; validate the antibody in your specific sample system.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider optimizing antigen retrieval and fixation/permeabilization protocols to ensure adequate epitope accessibility.
- As a rabbit polyclonal antibody, lot-to-lot consistency should be verified for critical quantitative assays; consider using appropriate positive and negative controls.
