Biological Background: POU4F2/BRN3B Function and Localization

  • POU4F2 (BRN3B) is a tissue-specific DNA-binding transcription factor that functions as both an activator and repressor of RNA polymerase II-dependent transcription, depending on the promoter context. It is involved in the development and differentiation of target cells. Related references: PMID:19266028 PMID:23805044
  • Plays a fundamental role in retinal ganglion cell (RGC) differentiation by binding to octamer motifs and forming a ternary complex with ISL1 and ISL2 to activate RGC-specific genes; it also inhibits DLX1 and DLX2 to prevent amacrine cell specification.
  • Interacts with multiple transcription factors: cooperates with TP53 to promote apoptosis of neuronal cells by activating BAX, acts as a coactivator for ESR1 on estrogen-responsive promoters, and antagonizes POU4F1’s transcriptional activity.
  • Primarily localizes to the nucleus, but can also be found in nuclear speckles and the cytoplasm.
  • Expressed in the brain and in the ganglion cell layer of the retina. Related references: PMID:7691107
  • Associated with apoptosis, alternative splicing, and developmental processes as indicated by UniProt keywords.

Experimental Guidance and Technical Tips

  • The immunogen covers the N‑terminal 100 amino acids, suggesting the antibody may recognize full‑length POU4F2 and possibly alternative isoforms.
  • For Western blot, a band around 43 kDa is expected; validate the band size in your specific sample type.
  • In immunofluorescence/ICC, confirm nuclear localization in RGC‑related or neuronal cell lines.
  • Cross‑reactivity with mouse is reported; consider verifying performance in mouse tissues or cells before use.
  • For ELISA, titrate the antibody to determine optimal working concentration.
