- POU4F2 (BRN3B) is a tissue-specific DNA-binding transcription factor that functions as both an activator and repressor of RNA polymerase II-dependent transcription, depending on the promoter context. - Plays a fundamental role in retinal ganglion cell (RGC) differentiation by binding to octamer motifs and forming a ternary complex with ISL1 and ISL2 to activate RGC-specific genes; it also inhibits DLX1 and DLX2 to prevent amacrine cell specification.
- Interacts with multiple transcription factors: cooperates with TP53 to promote apoptosis of neuronal cells by activating BAX, acts as a coactivator for ESR1 on estrogen-responsive promoters, and antagonizes POU4F1’s transcriptional activity.
