Biological Background: POU3F2 Function and Localization
- POU3F2 (also known as BRN2, OCT7, OTF-7, POUF3, N-Oct3) is a member of the POU domain class 3 transcription factor family, encoded by the POU3F2 gene.
- It binds to the consensus DNA sequence consisting of half-sites ('GCAT') and ('TAAT') separated by a variable spacer, regulating transcription of neuronal genes.
- Acts as a transcriptional activator in synergy with SOX4, SOX11, or SOX12 to promote gene expression.
- Together with ASCL1 and MYT1L, POU3F2 can reprogram fibroblasts into induced neuronal (iN) cells, acting downstream of ASCL1 to access open chromatin.
- Localized exclusively in the nucleus, consistent with its role as a transcription factor.
- The protein is subject to phosphorylation (as indicated by the keyword 'Phosphoprotein'), which may modulate its activity.
- Expression is restricted to the neuroectodermal lineage, underscoring its role in neuronal differentiation and neurogenesis.
- The theoretical molecular weight is approximately 46.9 kDa (UniProt: P20265), with observed migration at approximately 47 kDa by SDS-PAGE.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-68) of human POU3F2; validate reactivity in your target species and sample type.
- For Western blot, consider using whole-cell lysates from neuronal cell lines (e.g., SH-SY5Y) or brain tissue extracts. The predicted band is ~47 kDa; confirm with a positive control.
- For ELISA, titrate the antibody against the immunogen peptide or recombinant protein to determine optimal working dilution.
- As a polyclonal antibody, it may recognize multiple isoforms or degradation products; include appropriate controls.
- Cross-reactivity has been reported for human; if working with other species, verify sequence homology of the immunogen.
