Biological Background: POU2F1/OCT1 Function and Localization

  • POU2F1, also known as Oct-1, OTF-1, and NF-A1, is a ubiquitously expressed transcription factor belonging to the POU domain class 2 family. Alternative splicing generates multiple isoforms, including a lymphocyte-specific isoform 2.
  • It binds specifically to the octamer motif (5'-ATTTGCAT-3') and functions as a transcriptional activator for promoters of small nuclear RNAs (snRNA), histone H2B, and immunoglobulin genes.
  • POU2F1 modulates transcriptional transactivation by nuclear hormone receptors such as NR3C1 (glucocorticoid receptor), AR (androgen receptor), and PGR (progesterone receptor), integrating diverse signaling pathways.
  • During herpes simplex virus (HSV) infection, POU2F1 forms a multiprotein-DNA complex with the viral transactivator VP16 and host factor HCFC1, enabling transcription of viral immediate early genes, highlighting its role in host-virus interactions.
  • The protein localizes exclusively to the nucleus, consistent with its role as a DNA-binding transcription factor.
  • Key structural features include a POU-specific domain and a POU homeodomain that mediate DNA binding, and the protein is subject to phosphorylation, which may regulate its activity and interactions.
  • With a molecular weight of approximately 76 kDa, POU2F1 is widely detected across tissues, and its function is critical for both basal and inducible gene expression.

Experimental Guidance and Technical Tips

  • This antibody was raised against a recombinant protein fragment (aa 15-290) of human POU2F1/OCT1, encompassing the N-terminal region upstream of the POU domain. Verify reactivity in your target species and experimental system.
  • For Western blotting, a band around 76 kDa is expected; however, alternative splicing or post-translational modifications may produce additional bands. Optimize loading and blocking conditions.
  • ELISA applications may require optimization of coating antigen and antibody concentration; consider using the immunogen or a peptide for direct binding assays.
  • Cross-reactivity with rat has been reported, but performance in other species, including mouse, should be tested if needed.
  • Polyclonal antibodies may exhibit lot-to-lot variability; validate each lot in your specific application.
