- Serum paraoxonase/lactonase 3 (PON3) is a secreted enzyme encoded by the PON3 gene. It belongs to the paraoxonase family and has low activity toward organophosphates like paraxon and aromatic carboxylic acid esters.
- PON3 efficiently hydrolyzes lactones, including statin prodrugs such as lovastatin. It acts on aromatic lactones and 5- or 6-member ring lactones with aliphatic substituents, but not on simple lactones or those with polar substituents.
- The protein is secreted into the extracellular space, where it circulates in the blood and likely associates with high-density lipoproteins (HDLs), contributing to its anti-atherogenic properties.
- PON3 is a glycoprotein with a signal peptide, and it contains disulfide bonds and calcium-binding sites. Post-translational modifications include phosphorylation, and its enzymatic activity is metal-dependent.
