• Serum paraoxonase/lactonase 3 (PON3) is a secreted enzyme encoded by the PON3 gene. It belongs to the paraoxonase family and has low activity toward organophosphates like paraxon and aromatic carboxylic acid esters.
• PON3 efficiently hydrolyzes lactones, including statin prodrugs such as lovastatin. It acts on aromatic lactones and 5- or 6-member ring lactones with aliphatic substituents, but not on simple lactones or those with polar substituents.
