Biological Background: PON1 Function and Localization
- Serum paraoxonase/arylesterase 1 (PON1), also known as aromatic esterase 1, K-45, and serum aryldialkylphosphatase 1, is a secreted enzyme primarily associated with high-density lipoprotein (HDL) in plasma.
- PON1 hydrolyzes toxic metabolites of a broad spectrum of organophosphorus insecticides and can also hydrolyze lactones and aromatic carboxylic acid esters.
- The enzyme protects low-density lipoproteins (LDL) against oxidative modification, which is a critical step in the prevention of atheroma formation and cardiovascular disease.
- PON1 is expressed in the liver but is not detected in heart, brain, placenta, lung, skeletal muscle, kidney, or pancreas; its presence in plasma is tightly linked to HDL particles.
- The protein contains key structural features including calcium-binding sites, a disulfide bond, and glycosylation, and it has been extensively characterized by 3D-structure determination and direct protein sequencing.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 9–109 of human PON1 (NP_000437.3), which lies within the mature secreted protein after signal peptide cleavage; this region is expected to be conserved across human, mouse, and rat, supporting the observed cross-reactivity.
- For Western blot (WB) analysis, consider using liver tissue lysates or plasma/serum samples, as PON1 is highly expressed in the liver and secreted into the circulation; validate the antibody in your specific sample matrix.
- In ELISA applications, consider using recombinant PON1 protein as a standard and confirm detection sensitivity in human serum or plasma; the antibody's polyclonal nature may provide robust signal but requires careful optimization.
- Since PON1 is a secreted protein, if analyzing cellular extracts, ensure to include appropriate controls and consider potential limitations in detecting intracellular forms.
