Biological Background: POMGNT1 Function and Localization
- POMGNT1 (Protein O-linked-mannose beta-1,2-N-acetylglucosaminyltransferase 1) is a Golgi-resident glycosyltransferase, also designated GnT I.2, essential for the O-mannosyl glycan pathway.
- POMGNT1 catalyzes the transfer of N-acetylglucosamine (GlcNAc) to O-linked mannose residues on alpha-dystroglycan and other O-mannosylated proteins, forming the GlcNAc(β1-2)Man(α1-)O-Ser/Thr core. It is specific for α-linked terminal mannose and lacks MGAT3/4/5/7/8 activity (PubMed:11709191, PubMed:27493216, PubMed:28512129). Related references: PMID:11709191 PMID:27493216 PMID:28512129
- The gene undergoes alternative splicing, yielding multiple isoforms that may contribute to tissue-specific regulation of O-mannosylation.
- POMGNT1 is anchored in the Golgi apparatus membrane via a signal-anchor transmembrane domain, where it performs its glycosylation function.
- It is constitutively expressed, with relatively higher levels detected in spinal cord, lymph node, and trachea; strong expression is observed in astrocytes as well as immature and mature neurons.
- Mutations in POMGNT1 cause a spectrum of dystroglycanopathies, including congenital muscular dystrophy, limb-girdle muscular dystrophy (LGMD2O/LGMDR15), and retinitis pigmentosa.
- The protein contains structural features typical of glycosyltransferases: disulfide bonds, phosphorylation sites, and a manganese-dependent metal-binding domain.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 411–660 of human POMGNT1 (C-terminal region); the resulting polyclonal antibody may recognize multiple isoforms and is suitable for detecting the full-length protein (~75 kDa).
- For western blot, consider using protein lysates from human, mouse, or rat tissues with known POMGNT1 expression (e.g., brain, skeletal muscle) and validate with positive and negative controls.
- When performing ELISA, the antibody can be evaluated as a capture or detection reagent; include appropriate blocking and optimize dilution factors for your specific assay format.
- As with any polyclonal antibody, lot-to-lot consistency should be verified, and further dilution optimization is recommended for each application and sample type.
