- POLR2D, also known as RPB4, RPB16, or DNA-directed RNA polymerase II subunit D, is a core subunit of RNA polymerase II (Pol II), responsible for synthesizing mRNA precursors and non-coding RNAs.
- It forms a stable subcomplex with POLR2G/RPB7 that binds to a specific pocket formed by the large subunits RPB1, RPB2, and RPB6 at the base of the Pol II clamp element.
- The POLR2D/RPB4-POLR2G/RPB7 subcomplex locks the clamp in a closed conformation via POLR2G/RPB7, preventing double-stranded DNA from entering the active site cleft and thereby regulating transcription initiation.
- This subcomplex also possesses single-stranded DNA and RNA binding activity, contributing to polymerase processivity and transcript stabilization.
