Biological Background: POLD2 Function and Localization
- POLD2 encodes DNA polymerase delta subunit 2 (also known as p50), an accessory component of both the DNA polymerase delta and zeta complexes, playing a role in high fidelity genome replication (including lagging strand synthesis) and repair. - Exists in two forms: Pol-delta3 (lacking POLD4) and Pol-delta4 (containing POLD4); Pol-delta3 exhibits higher proofreading activity and reduced strand displacement, potentially better suited for Okazaki fragment processing, while Pol-delta4 stalls upon encountering 5'-blocking oligonucleotides. - Along with DNA polymerase kappa, DNA polymerase delta carries out approximately half of nucleotide excision repair (NER) synthesis following UV irradiation. Related references: PMID:20227374
