Biological Background: POGLUT1 Function and Localization
- POGLUT1 is a dual specificity glycosyltransferase that transfers glucose and xylose from UDP-sugars to serine residues within the consensus motif C-X-S-X-P-C on epidermal growth factor (EGF) repeats of target proteins. Related references: PMID:21081508, PMID:21490058, PMID:21949356
- It acts as a positive regulator of Notch signaling by mediating O-glucosylation of Notch extracellular EGF repeats, a modification critical for proper muscle development. Related references: PMID:27807076
- The enzyme localizes to the endoplasmic reticulum lumen, where it modifies nascent EGF repeat-containing proteins before trafficking.
- It is broadly expressed in adult tissues, with highest abundance in liver, and also detected in brain, heart, skeletal muscle, spleen, kidney, placenta, lung, and peripheral blood leukocytes. At the protein level, it is prominent in the upper epidermal layers.
- Mutations in POGLUT1 are associated with limb-girdle muscular dystrophy (LGMD2Z / LGMDR21), highlighting its role in muscle maintenance.
- Additional known targets include CRB2, F7, and F9, and its glycosylation of CRB2 is required for early development and gastrulation.
Experimental Guidance and Technical Tips
- For Western blot, expect a band around 46 kDa. Use reducing conditions and load appropriate protein amounts from human, mouse, or rat lysates. Optimize blocking and antibody dilution for specific signal.
- In IF/ICC, fix cells with paraformaldehyde, permeabilize, and validate co-localization with an ER marker such as calreticulin or PDI.
- For ELISA, employ recombinant POGLUT1 protein or peptide fragments as coating antigen. Pair with appropriate secondary antibody and chromogenic substrate; include standard curves.
- The immunogen corresponds to amino acids 160–280 of human POGLUT1; therefore, the antibody recognizes an internal region. Cross-reactivity with mouse and rat is expected based on sequence homology.
