  • Podocalyxin (PODXL), also known as GCTM-2 antigen, Gp200, or podocalyxin-like protein 1 (PCLP-1), is a single-pass type I transmembrane glycoprotein belonging to the CD34 family. It is primarily expressed on glomerular podocytes and has roles in cell adhesion and morphology.
  • Functions as both an anti-adhesive and pro-adhesive molecule: maintains an open filtration pathway between podocyte foot processes via charge repulsion, while in other contexts, it enhances integrin-dependent cell adhesion, migration, and cell-cell contacts.
  • Induces the formation of apical actin-dependent microvilli and participates in establishing epithelial polarity and apical lumen formation during renal tubulogenesis.
  • Contributes to cancer progression by promoting cell migration and invasion through interaction with the actin-binding protein EZR, leading to activation of MAPK and PI3K/AKT signaling pathways.
  • Localizes to the apical cell membrane and is enriched in membrane projections such as microvilli, lamellipodia, filopodia, and ruffles, as well as membrane rafts.
  • The protein undergoes post-translational modifications including phosphorylation and glycosylation, which may regulate its function. Alternative splicing generates multiple isoforms.
