Biological Background: Podoplanin Function and Localization
- Podoplanin (Aggrus, Gp36, PA2.26 antigen) is a small transmembrane glycoprotein of approximately 17 kDa (calculated molecular weight 16.7 kDa), with extensive O-glycosylation and sialic acid modifications.
- It mediates cell migration and adhesion through interactions with partners such as CLEC1B, CD9, MSN, EZR, and CD44; during development, it is critical for blood/lymphatic vessel separation by triggering platelet activation via CLEC1B binding. - For Western blot (WB), the predicted molecular weight is approximately 17 kDa, but extensive O-glycosylation can result in higher apparent molecular weights (40–45 kDa). Validate band specificity in the relevant sample system.
- In immunohistochemistry (IHC-P), antigen retrieval methods (e.g., heat-induced epitope retrieval) and tissue-specific fixation conditions should be optimized to ensure specific staining.
- The antibody is listed as cross-reactive with mouse and rat Podoplanin; however, verification in the intended model system is recommended.
