Biological Background: Podoplanin Function and Localization

  • Podoplanin (PDPN, also known as Aggrus, GP36, T1A) is a type I transmembrane glycoprotein with extracellular, transmembrane, and cytoplasmic domains. It undergoes alternative splicing and is heavily sialylated.
  • Through binding to CLEC1B, podoplanin triggers platelet activation and aggregation, key to blood/lymphatic vessel separation (PubMed:14522983, PubMed:15231832, PubMed:17222411). Interaction with CD9 attenuates this aggregation (PubMed:18541721). Related references: PMID:14522983 PMID:15231832 PMID:17222411 PMID:18541721
  • Interaction with MSN or EZR promotes epithelial-mesenchymal transition (EMT), leading to ERZ phosphorylation and RHOA activation, thereby enhancing cell migration and invasiveness (PubMed:17046996, PubMed:21376833). Related references: PMID:17046996 PMID:21376833
  • Podoplanin binding to CD44 promotes directional cell migration in epithelial and tumor cells (PubMed:20962267). It also controls invadopodia stability and extracellular matrix degradation via the RHOC-ROCK-LIMK-CFL1 pathway (PubMed:25486435). Related references: PMID:20962267 PMID:25486435
  • In lymph nodes, podoplanin regulates fibroblastic reticular cell adhesion and contractility through ERM proteins and MYL9 activation; CLEC1B engagement induces relaxation (By similarity).
  • Podoplanin localizes primarily to the plasma membrane and is enriched in membrane projections: lamellipodia, filopodia, microvilli, ruffles, and invadopodia. It also traffics to membrane rafts and shows apical/basolateral polarity.
  • Highly expressed in placenta, lung, skeletal muscle, and brain; upregulated in colorectal tumors and some oral squamous cell carcinomas.
  • In keratinocytes, podoplanin expression leads to elongated morphology, actin cytoskeleton reorganization, increased motility, and decreased cell adhesion (PubMed:15515019). Related references: PMID:15515019

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 23-131, a region within the extracellular domain; this design likely targets epitopes accessible in native and denatured conditions, supporting WB and IHC-P applications.
  • For WB, the predicted molecular weight is ~17 kDa; however, extensive O-glycosylation and sialylation may cause bands to appear at 36–44 kDa or higher under reducing conditions. Validate using appropriate positive controls such as lung or placenta lysates.
  • For IHC-P, consider antigen retrieval and validate staining in known positive tissues (e.g., placenta, lung, tumor sections) with appropriate negative controls.
  • Cross-reactivity with mouse and rat has been indicated; however, verify sequence homology and expected band sizes across species before use in multi-species experiments.
  • For ELISA, the recombinant immunogen can serve as a capture standard; test coating conditions and ensure detection antibody compatibility.
