Biological Background: Podoplanin Function and Localization

  • Podoplanin (PDPN), also known as Gp38, OTS-8, E11, and T1alpha, is a type I transmembrane sialoglycoprotein of approximately 18 kDa expressed as an alternative PA2.26 antigen.
  • It mediates cell migration and adhesion through diverse partners: binding CLEC1B triggers platelet activation/aggregation, crucial for blood-lymphatic vessel separation during development (PubMed:14522983, PubMed:15231832, PubMed:17616532). Related references: PMID:14522983 PMID:15231832 PMID:17616532
  • Conversely, interaction with CD9 attenuates platelet aggregation and metastasis; interactions with MSN or EZR promote epithelial-mesenchymal transition (EMT) via ERZ phosphorylation and RHOA activation, increasing invasiveness.
  • In lymph nodes, PDPN controls fibroblastic reticular cell (FRC) adhesion and contraction by maintaining activated ERM proteins and MYL9; CLEC1B engagement promotes relaxation (PubMed:25347465). Related references: PMID:25347465
  • In keratinocytes, ectopic expression induces elongated morphology, actin reorganization, increased motility, and decreased adhesion (PubMed:10574709). Related references: PMID:10574709
  • Subcellularly, it localizes to plasma membrane domains including lamellipodia, filopodia, microvilli, ruffles, membrane rafts, apical and basolateral membranes, and invadopodia, consistent with its role in cell migration and matrix degradation.
  • Tissue distribution is highest in lung and brain, with lower levels in kidney, stomach, liver, spleen, esophagus; expressed in lymphatic endothelia, epithelial cells of choroid plexus, glomerulus, alveoli, and stromal cells of lymphoid tissues; up-regulated in colon carcinomas.
  • It is required for normal lung cell proliferation and alveolus formation at birth (PubMed:12654292). Related references: PMID:12654292
  • Additional keywords: cell junction, developmental protein, glycoprotein, sialic acid; it does not function as a water channel or folate transporter (PubMed:12032185). Related references: PMID:12032185

Experimental Guidance and Technical Tips

  • For Western blotting, target the expected band at approximately 18 kDa, though glycosylation may cause a higher apparent molecular weight; include appropriate positive controls (e.g., lung or lymphatic tissue lysates).
  • In immunohistochemistry and immunofluorescence, consider using tissues with known high Podoplanin expression (e.g., lung, lymphatic vessels) or relevant cell lines; antigen retrieval may be necessary for optimal staining.
  • As a rabbit monoclonal antibody, consistent lot-to-lot performance is expected; validate in your specific experimental system to confirm suitability for quantitative assays.
