Biological Background: RNA-binding protein PNO1 Function and Localization
- RNA-binding protein PNO1 (Partner of NOB1) is a component of the small subunit (SSU) processome, the earliest precursor of the small eukaryotic ribosomal subunit. It participates in ribosome biogenesis, facilitating pre-rRNA folding, modifications, and cleavage.
- PNO1 positively regulates dimethylation of two adjacent adenosines in a conserved hairpin near the 3′-end of 18S rRNA, a critical modification for ribosomal function. Related references: PMID:25851604
- As part of the SSU processome, PNO1 works with ribosomal proteins and RNA chaperones to ensure proper assembly and targeted degradation of pre-ribosomal RNA by the RNA exosome. Related references: PMID:34516797
- PNO1 localizes primarily to the nucleolus, the site of ribosomal RNA synthesis and assembly.
- Expression is observed in liver, lung, spleen, and kidney, with weaker expression in thymus, testis, and ovary. It is weakly or not expressed in heart, brain, skeletal muscle, placenta, pancreas, prostate, small intestine, colon, and peripheral blood leukocytes.
- The protein contains RNA-binding domains and undergoes acetylation, as identified by proteomics (UniProt keywords: 3D-structure, Acetylation, Nucleus, Proteomics identification, Reference proteome, RNA-binding).
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 1–252 of human PNO1, providing recognition of the full-length protein. Expect a band around 28 kDa in Western blot.
- For WB, consider using positive control lysates from liver, lung, spleen, or kidney, where PNO1 expression is relatively higher.
- For ELISA, the recombinant immunogen may be used as a positive control to assess antibody binding.
- Validate antibody specificity with siRNA-mediated knockdown of PNO1 or by pre-incubating the antibody with the immunogen peptide.
- Although validated for human and rat, cross-reactivity with other species may be possible but requires verification.
