Biological Background: PNLIPRP2 (PLRP2) Function and Localization

  • PNLIPRP2, also designated pancreatic lipase-related protein 2 (PLRP2), is alternatively known as cytotoxic T lymphocyte lipase, galactolipase, and triacylglycerol lipase.
  • Functions primarily as a lipase that hydrolyzes triglycerides and galactosylglycerides, with activity against short-, medium-, and long-chain fatty acyls without positional specificity. It can completely deacylate triacylglycerols and exhibits low phospholipase activity. Related references: PMID:15287741, PMID:17401110, PMID:18702514, PMID:21865348
  • In neonates, plays a major role in pancreatic digestion of dietary fats, particularly milk fat globules enriched in long-chain triglycerides. Related references: PMID:19824014, PMID:21652702, PMID:23732775
  • In adults, likely functions mainly as a galactolipase and monoacylglycerol lipase, hydrolyzing monogalactosyldiglycerols (MGDG) and digalactosyldiacylglycerols (DGDG) from plant-based diets, releasing long-chain polyunsaturated fatty acids. Related references: PMID:15287741, PMID:17401110, PMID:18702514, PMID:20083229
  • May act together with gastric lipase (LIPF) to hydrolyze partially digested triglycerides. Related references: PMID:23732775
  • In cytotoxic T cells, contributes to perforin-dependent cell lysis (by similarity). In neurons, required for the localization of the phospholipid 1-oleoyl-2-palmitoyl-PC (OPPC) to neurite tips, promoting surface expression of the dopamine transporter SLC6A3/DAT by facilitating fusion of transport vesicles with the plasma membrane (by similarity).
  • Found as a secreted protein, associated with zymogen granule membranes, and localized to cell projections including neuron projections.
  • Highly expressed in the pancreas; contains structural features such as calcium-binding sites, disulfide bonds, and glycosylation motifs.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 301–470 of human PNLIPRP2, covering a region within the mature protein. This likely generates antibodies recognizing multiple epitopes, suitable for detecting endogenous PNLIPRP2.
  • For Western blotting (WB), expect a band near 52 kDa in reduced samples; validate detection in your specific tissue or cell lysate, and titrate antibody concentrations.
  • In immunofluorescence (IF-P), PNLIPRP2 localizes to secretory vesicles and neuronal projections. Consider optimizing fixation and permeabilization to detect membrane-associated pools.
  • For ELISA applications, pair with a suitable capture/detection antibody. Validate with recombinant PNLIPRP2 or cell supernatants to assess sensitivity.
  • Cross-reactivity with mouse and rat enables translational studies, but verify performance in your model system as epitope conservation varies.
