Biological Background: PNLIPRP1 Function and Localization
- PNLIPRP1 (also known as PLRP1) is an inactive pancreatic lipase-related protein 1, belonging to the AB hydrolase superfamily.
- Unlike active lipases, PNLIPRP1 exhibits very low or undetectable catalytic activity towards triglycerides, diglycerides, phosphatidylcholine, galactolipids, or cholesterol esters (By similarity).
- In vitro, PNLIPRP1 is unable to hydrolyze milk fat, indicating its lack of lipolytic function under physiological conditions. Related references: PMID:19824014
- The PNLIPRP1 gene is subject to alternative splicing, potentially generating multiple isoforms (UniProt keyword).
- PNLIPRP1 is a secreted protein, consistent with its role in the pancreatic exocrine function.
- Tissue expression is predominantly in the pancreas, where it is produced alongside other pancreatic lipases.
- The protein structure includes features such as calcium-binding sites, disulfide bonds, and a signal peptide, characteristic of secreted lipases (Keywords: Calcium, Disulfide bond, Signal).
- While its biological role remains unclear, PNLIPRP1 may serve regulatory or structural functions in lipid metabolism.
Experimental Guidance and Technical Tips
- When performing Western blot, a band at approximately 52 kDa corresponding to PNLIPRP1 may be observed. Validate with appropriate positive controls such as pancreatic lysates.
- For immunohistochemistry, pancreas tissue sections are recommended due to the protein's tissue-specific expression.
- The immunogen covers the C-terminal region (aa 298–467); therefore, the antibody may detect full-length and possibly cleaved forms.
- Antibody cross-reactivity with mouse and rat has been reported; consider sequence alignment for other species to anticipate potential cross-detection.
- Standard blocking and detection protocols for rabbit polyclonal antibodies are applicable; optimization of antibody dilution and incubation conditions is advised for each application.
