Biological Background: PMS2 Function and Localization
- PMS2 (PMS1 protein homolog 2), also termed MLH4, PMS-2, PMSL2, HNPCC4, LYNCH4, or MMRCS4, is a 96 kDa mismatch repair endonuclease essential for maintaining genomic stability.
- It is a key component of the post-replicative DNA mismatch repair (MMR) system, forming a heterodimer with MLH1 (MutL alpha). This complex is recruited to mismatch sites by MutS alpha (MSH2-MSH6) or MutS beta (MSH2-MSH3) upon recognition of base-base mismatches or insertion/deletion loops. - Upon recruitment, PMS2 endonuclease activity introduces single-strand breaks near the mismatch, creating entry points for EXO1 exonuclease to degrade the error-containing strand. DNA methylation likely directs cleavage to the newly synthesized strand, ensuring strand-specific repair.
- PMS2 also possesses ATPase activity; however, ATP hydrolysis may not be required for proficient mismatch repair as gross conformational changes are absent.
- Beyond mismatch repair, PMS2 is implicated in DNA damage signaling, promoting cell cycle arrest and apoptosis in response to severe DNA lesions.
- The protein localizes predominantly in the nucleus, consistent with its DNA repair functions, and undergoes alternative splicing, leading to multiple isoforms.
- Mutations in PMS2 are linked to hereditary nonpolyposis colorectal cancer (HNPCC, Lynch syndrome) and other mismatch repair cancer syndromes, classifying it as a tumor suppressor.

- For Western blotting, the target protein runs at approximately 96 kDa; consider using fresh lysates from human colorectal cell lines (e.g., HCT116, SW480) as positive controls and validate knockout/knockdown models to confirm band specificity.
- For ELISA applications, optimization of coating antigen and antibody concentration is recommended; test with recombinant human PMS2 protein to assess sensitivity and linearity.
- Since the antibody is produced against recombinant human PMS2 protein, cross-reactivity with other species is not expected; if attempting other species, perform sequence alignment and validation.
