Biological Background: PMS1 Function and Localization
- PMS1 (PMS1 protein homolog 1), also known as DNA mismatch repair protein PMS1, is encoded by the PMS1 gene (Gene ID: 5378). Alternative splicing generates multiple isoforms, with the canonical form having a molecular weight of approximately 106 kDa (UniProt: 105.83 kDa).
- PMS1 is probably involved in the repair of DNA mismatches, a critical process for maintaining genomic stability by correcting base-base and insertion-deletion mispairs that arise during DNA replication.
- As a component of the mismatch repair system, PMS1 functions in concert with other MMR proteins and is associated with hereditary nonpolyposis colorectal cancer (HNPCC) when mutated.
- PMS1 localizes predominantly to the nucleus, where it accesses DNA to perform its repair functions.
- The protein contains DNA-binding domains, consistent with its role in recognizing and processing mismatched DNA segments.
- PMS1 is classified as a tumor suppressor, and its dysfunction is linked to DNA damage and tumorigenesis.
Experimental Guidance and Technical Tips
- The polyclonal antibody is raised against a recombinant immunogen corresponding to the C-terminal region (aa 405-619) of human PMS1. For Western blot, consider using nuclear lysates from human cell lines, as PMS1 is a nuclear protein with an expected band around 106 kDa.
- For ELISA, the antibody may recognize the immunogen region; consider coating plates with the recombinant protein or nuclear extracts from human samples.
- Cross-reactivity is reported for human; for other species, sequence alignment of the immunogen region is recommended to predict potential cross-reactivity.
- Validation in mismatch repair-proficient and -deficient cell models may provide additional specificity controls.
