Biological Background: PML Function and Localization
-
PML (Promyelocytic leukemia protein), also designated TRIM19, MYL, and RNF71, is a member of the tripartite motif (TRIM) family and functions as an E3 SUMO-protein ligase.

PMID:20972456 PMID:28250117 -
PML serves as the scaffold of PML nuclear bodies (PML-NBs), subnuclear organelles that regulate key processes such as tumor suppression, transcriptional control, apoptosis, senescence, and DNA damage repair.
-
It suppresses mRNA export by inhibiting the cap-binding protein EIF4E, reducing its affinity for the 5' 7-methylguanosine cap of target mRNAs.

PMID:11500381 PMID:11575918 PMID:18391071 -
Multiple isoforms generated by alternative splicing display specialized functions: PML-4 represses TBX2 during senescence and modulates p53 stability, while PML-6 specifically inhibits the tetrameric form of PKM.
-
PML exhibits broad antiviral activity, restricting DNA and RNA viruses such as herpesviruses, influenza A, rabies, and poliovirus via capsid sequestration, polymerase trapping, or apoptosis induction.
-
PML localizes to the nucleus, nucleoplasm, PML bodies, nucleolus, and also dynamically associates with cytoplasmic structures such as the endoplasmic reticulum and endosomes, enabling its multitasking roles.
-
Post-translational modifications, including acetylation, phosphorylation, and SUMOylation, tightly regulate PML’s activity and its interactions within PML-NBs.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 571–611 of human PML (isoform NP_150241.2); consult isoform alignment to anticipate potential cross-reactivity among splicing variants.
- For Western blot, a band near 98 kDa is expected, but additional bands from other isoforms or post‑translational modifications may appear; validate using known positive and negative controls.
- For immunohistochemistry (IHC-P), consider testing on tissues known to express PML, such as spleen, thymus, or tumor biopsies, and include appropriate controls.
- As a polyclonal antibody, lot‑to‑lot consistency is important; perform titration experiments to determine the optimal working dilution for each application.
