Biological Background: PLZF Function and Localization
- PLZF (Promyelocytic leukemia zinc finger protein), also known as Zinc finger and BTB domain-containing protein 16 (ZBTB16) or Zinc finger protein 145 (ZNF145), is a transcriptional repressor encoded by the ZBTB16 gene.
- It functions as a transcriptional repressor, partly by recruiting histone deacetylases to target gene promoters (PubMed:10688654, PubMed:24359566). Related references: PMID:10688654 PMID:24359566
- PLZF is a substrate-recognition component of an E3 ubiquitin-protein ligase complex, mediating ubiquitination and proteasomal degradation of target proteins (PubMed:14528312). Related references: PMID:14528312
- The protein predominantly localizes to the nucleus, particularly in nuclear bodies, consistent with its role in transcriptional regulation.
- Within the hematopoietic system, PLZF is expressed in bone marrow, early myeloid cell lines, and peripheral blood mononuclear cells, with additional expression in ovary, kidney, and lung.
- PLZF contains zinc finger and BTB/POZ domains, classifying it as a metal-binding, DNA-binding transcription factor; it undergoes phosphorylation and is linked to the ubiquitin conjugation pathway.
- Alternative splicing generates multiple isoforms, and chromosomal rearrangements involving ZBTB16 are associated with acute promyelocytic leukemia.
Experimental Guidance and Technical Tips
- For Western blotting, PLZF has a predicted molecular weight of ~74 kDa. Use an appropriate gel percentage (e.g., 10%) and optimize transfer conditions for efficient detection.
- The immunogen (amino acids 199–300) resides in the protein’s central region. Check sequence conservation across species to anticipate potential cross-reactivity.
- This antibody is compatible with IP and IF-P; for nuclear antigens, consider proper fixation and permeabilization protocols to ensure nuclear access.
- Reported cross-reactivity with mouse and rat PLZF should be validated in the target sample system, as recognition may vary with endogenous expression levels.
- In ELISA, preliminary checkerboard titrations with a matched detection antibody are recommended to establish optimal working concentrations.
