Biological Background: PLXNA3 Function and Localization
- PLXNA3, also known as Plexin-A3, Plexin-4, or Semaphorin receptor SEX, is a single-pass transmembrane receptor belonging to the plexin family.
- It acts as a coreceptor for class 3 semaphorins SEMA3A and SEMA3F, forming signaling complexes with neuropilins to mediate cytoskeletal reorganization.
- PLXNA3 is essential for axon guidance and neuronal migration in the developing nervous system, regulating sympathetic neuron migration and dendritic spine morphology in pyramidal neurons.
- The protein may also be involved in semaphorin-mediated programmed cell death during neurodevelopment.
- PLXNA3 is localized at the cell membrane, with a large N-terminal extracellular region containing sema and PSI domains, a transmembrane helix, and a cytoplasmic tail required for downstream signaling.
- It undergoes glycosylation, disulfide bond formation, and phosphorylation, consistent with its role as a cell-surface receptor involved in signal transduction.
Experimental Guidance and Technical Tips
- Based on the immunogen sequence (amino acids 950–1150), the antibody is expected to recognize the cytoplasmic domain of human PLXNA3.
- For Western blot (WB), a band at approximately ~208 kDa is anticipated; due to the high molecular weight, consider using gradient gels and extended transfer times for optimal resolution and blotting.
- Validate the antibody in relevant human cell lines or tissues known to express PLXNA3 to confirm specificity.
- For ELISA, the antibody can be employed in both direct and capture formats; optimization of coating concentrations and detection systems is recommended.
- Cross-reactivity with human PLXNA3 is specified; for samples from other species, sequence homology should be examined, and experimental validation is advised.
