Biological Background: PLTP Function and Localization

  • Phospholipid transfer protein (PLTP), also known as Lipid transfer protein II, is a secreted protein that plays a central role in lipid metabolism.
  • PLTP mediates the transfer of phospholipids and free cholesterol from triglyceride-rich lipoproteins (VLDL and LDL) to high-density lipoproteins (HDL), and facilitates the exchange of phospholipids between triglyceride-rich lipoproteins themselves.
    Related references: PMID:11013307, PMID:19321130, PMID:21515415
  • It transfers a broad spectrum of lipid molecules, including diacylglycerol, phosphatidic acid, sphingomyelin, phosphatidylcholine, phosphatidylinositol, phosphatidylglycerol, cerebroside, and phosphatidyl ethanolamine.
    Related references: PMID:9132017
  • PLTP is critical for HDL remodeling, modulating the size and composition of HDL particles.
    Related references: PMID:29883800
  • It also contributes to reverse cholesterol transport by facilitating the uptake of cholesterol from peripheral cells for hepatic degradation and excretion.
    Related references: PMID:21736953
  • Two distinct forms of PLTP are present in plasma: an active form capable of transferring phosphatidylcholine to HDL, and an inactive form lacking this activity.
    Related references: PMID:11013307
  • Subcellularly, PLTP is secreted but also detected in the nucleus. It is widely expressed, with highest levels in ovary, thymus, and placenta, and moderate expression in pancreas, small intestine, testis, lung, and prostate; expressed in cortical neurons.
  • Post-translational modifications include disulfide bonds and glycosylation, and the gene undergoes alternative splicing.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 142-441 of human PLTP, a region that may be present in both active and inactive isoforms; this could allow detection of total PLTP in biological samples.
  • For Western blotting under reducing conditions, consider using plasma or lysates from tissues with high PLTP expression (e.g., ovary, thymus, placenta) and validate the antibody in the relevant sample system.
  • For ELISA applications, optimize coating conditions and blocking buffers to minimize nonspecific binding, and include appropriate negative controls.
  • Cross-reactivity with mouse PLTP enables potential application in mouse models of lipid metabolism and cardiovascular disease; however, performance should be empirically verified.
