Biological Background: PLSCR3 Function and Localization
- PLSCR3 catalyzes calcium-induced, ATP-independent bidirectional phospholipid scrambling (flip-flop) at the mitochondrial membrane, equilibrating phospholipids between the inner and outer leaflets. Related references: PMID:14573790 PMID:17226776 PMID:18358005
- It plays a critical role in mitochondrial respiratory function, morphology, and apoptotic signaling by modulating cardiolipin distribution. Related references: PMID:12649167 PMID:14573790 PMID:17226776
- PLSCR3 mediates the translocation of cardiolipin from the inner mitochondrial membrane to the outer membrane, enhancing t-Bid-induced cytochrome c release and apoptosis. Related references: PMID:14573790 PMID:17226776 PMID:18358005
- It regulates both de novo biosynthesis and resynthesis of cardiolipin, maintaining mitochondrial membrane integrity. Related references: PMID:16939411
- PLSCR3 is primarily localized to the mitochondrial inner membrane but can also be found in the nucleus.
- According to UniProt, PLSCR3 is detected in a wide range of tissues including heart, placenta, lung, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, uterus, small intestine, and peripheral blood lymphocytes, while expression is not detected in testis and brain.
- PLSCR3 is known to be palmitoylated and phosphorylated, modifications that may regulate its activity and localization.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human PLSCR3 (aa 1-295), which may allow detection of both endogenous and overexpressed protein.
- For Western blotting, using mitochondrial-enriched lysates or whole-cell lysates from tissues with high PLSCR3 expression (e.g., heart, placenta) is recommended to improve signal intensity.
- In immunofluorescence/immunocytochemistry (IF/ICC), include mitochondrial co-staining to confirm specific localization.
- Due to the polyclonal nature, antibody specificity should be validated with appropriate controls (e.g., knockdown or knockout cell models) in the relevant sample system.
- For ELISA applications, pairing with a specific capture antibody may be necessary; optimization of coating conditions and detection strategy is recommended.
