Biological Background: PLOD3/LH3 Function and Localization
- PLOD3 (also known as LH3) is a multifunctional enzyme that catalyzes post-translational modifications of lysine residues in procollagen, including hydroxylation, galactosylation, and glucosylation, which are critical for collagen stability and basement membrane integrity. Related references: PMID:11956192 PMID:12475640 PMID:18298658
- It redundantly catalyzes the formation of hydroxylysine in -Xaa-Lys-Gly- motifs of collagens. Related references: PMID:11956192 PMID:12475640 PMID:18298658
- PLOD3 also transfers galactose onto hydroxylysine residues, forming galactosyl hydroxylysine. Related references: PMID:12475640 PMID:18298658 PMID:18834968
- Its essential role lies in the subsequent glucosyltransferase activity, producing 1,2-glucosylgalactosyl-5-hydroxylysine residues. Related references: PMID:10934207 PMID:11896059 PMID:11956192
- PLOD3 is required for normal biosynthesis and secretion of type IV collagens and basement membrane formation. Related references: PMID:18834968
- It localizes primarily to the rough endoplasmic reticulum, ER lumen, and ER membrane, and can be secreted into the extracellular space.
- The protein is ubiquitously expressed, with relatively higher levels in heart, placenta, and pancreas, and lower in lung, liver, and skeletal muscle. Related references: PMID:9724729 PMID:9582318
- PLOD3 is a glycosylated, metal-binding (iron and manganese) member of the dioxygenase and glycosyltransferase families, and mutations are associated with disease.
- It also hydroxylates and glycosylates the collagen-like domain of MBL1, expanding its substrate specificity. Related references: PMID:25419660
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 25-306 of human PLOD3, covering a region likely accessible to antibody binding. For Western blot, consider using reducing conditions to detect the 85kDa band, and validate with lysates from human and mouse cell lines known to express PLOD3 (e.g., heart, placenta).
- The antibody is validated for ELISA; for sandwich ELISA applications, pairing with another clone recognizing a distinct epitope may improve sensitivity. Cross-reactivity with mouse suggests utility in rodent model studies, but cross-reactivity with other species should be verified experimentally.
- Since PLOD3 is an ER-resident and secreted protein, sample preparation for WB may benefit from subcellular fractionation or enrichment strategies, particularly for samples with low endogenous expression.
