- PLOD2 encodes procollagen-lysine,2-oxoglutarate 5-dioxygenase 2, also known as lysyl hydroxylase 2 (LH2), an enzyme belonging to the 2-oxoglutarate-dependent dioxygenase family.
- The enzyme catalyzes the hydroxylation of lysine residues in Xaa-Lys-Gly sequences of collagen, forming hydroxylysine. These modified residues are critical for the attachment of carbohydrate units and for the stability of intermolecular collagen cross-links.
- PLOD2 localizes to the rough endoplasmic reticulum membrane and the cytoplasm, where it exerts its enzymatic function during procollagen processing.
- Two main isoforms exist due to alternative splicing; isoform 2 is selectively expressed in skin, lung, dura, and aorta, while both isoforms are broadly detected in other tissues.
- The protein is highly expressed in pancreas and muscle, consistent with the high collagen turnover in these tissues.
