- LH2 catalyzes the hydroxylation of lysine residues in -Xaa-Lys-Gly- sequences within collagen, forming hydroxylysine; these hydroxylysines serve as attachment sites for carbohydrates and are critical for the stability of intermolecular collagen cross-links.
- The enzyme is localized to the rough endoplasmic reticulum membrane and cytoplasm, where co-translational collagen modification occurs.
- Tissue expression is highest in pancreas and muscle; isoforms 1 and 2 are broadly expressed, with isoform 2 specifically detected in skin, lung, dura, and aorta.
